What is Trans-Polydatin 98%?

Polydatin (piceid), an active ingredient extracted from Polygonum cuspidatum, is a stilbenoid glucoside of resveratrol. A sugar molecule is jointed to the resveratrol structure, making polydatin more bioactive and powerful in antioxidant and anti-aging potency.

molecular structure comparison of polydatin and resveratrol

Resveratrol includes trans-resveratrol and cis-resveratrol. Like resveratrol, there are two kinds of polydatin derivatives: trans-polydatin and cis-polydatin. The bioactivity of trans-isomers is much higher than that of cis-isomers. There are two CAS number available for polydatin, one is 65914-17-2 from Sigma-Aldrich, and the other is 27208-80-6 from Wikipedia (as piceid). Scientific literatures, references, documents use piceid more often while in dietary supplement industry polydatin is preferred. If not specified, all the polydatin and piceid mentioned below will be trans-polydatin.

Botanical sources of polydatin extract

Except for the extraction from root and rhizome of polygonum cuspidatum (Japanese knotweed extract
), transpolydatin is also detected in grape, peanut, hop cones, red wines, hop pellets, cocoa-containing products, chocolate products and many daily diets. It can also be found in the bark of Picea sitchensis. However, polydatin is present at a much higher content (2% polydatin) in dried root of polygonum cuspidatum than in wine and any other sources.

Market status of polydatin sales

Polydatin has been the research object in China since 1970s. Its research began even earlier than resveratrol itself. However, R&D of polydatin takes a long period and its main applications are focused on pharmaceutical purposes as an analgesic, anti-pyretic, diuretic and expectorant over the past decades in domestic market of China. Polygonum cuspidatum extract standardized to 90% polydatin is the main specification for domestic markets.

With the increasing popularity of resveratrol in the international supplement market since 2006, polydatin comes to the focus of global dietary supplements. Polydatin is a precursor of resveratrol, and most importantly, it is water soluble. Resveratrol is not soluble, and it is only available in the forms of capsules. In addition, the specification of polydatin is 98% for nutritional supplements formula. Polydatin also begins to use in skincare cosmetics for its antioxidant and anti-aging prosperities. It is another trend for polydatin development in the next decade.

Polydatin VS Resveratrol (conversion and comparison)
Trans-resveratrol is one of the major metabolites of polydatin. After oral administration in humans, Polydatin (Piceid), 3,4,5-trihydroxystilbene-3-beta-D-glucoside, is hydrolyzed into trans-resveratrol, trans-resveratrol-3-O-glucuronide in the small intestine. As a result, polydatin dramatically increases the concentration and half life of resveratrol and its other metabolites in blood plasma and tissues, making resveratrol over 1,000 times more concentrated in the tissues of animals than is the case with the same metabolites of resveratrol.

Once the polydatin enters the bloodstream the sugar molecule is cleaved off, leaving the resveratrol. Polydatin allows resveratrol to skip phase I metabolism and enter into the blood stream more readily. This in turn means that the glucoside of resveratrol is absorbed at a different rate to standard trans-resveratrol, thereby effectively increasing the half-life and the potency of the resveratrol, which provides a bioavailability of polydatin 2x to 5x longer than resveratrol.

Mechanism of action of polydatin
How does polydatin work? Let’s first look at the relation between polydatin and resveratrol first. Polydatin is a pre-cursor to Resveratrol and activates Sirt1. Sirt1, or Sirtuin 1(silent mating type information regulation 2 homolog), is the human anti-aging Gene. Activation of SIRT1 makes your cells mitochondria (the powerhouse of the cell) more efficient and provides benefits to cellular energy production in bone cells, muscle cells, immune cells, etc

It shows increased bioavailability over Resveratrol because polydatin is more resistant to enzymatic oxidation than resveratrol, polydatin is soluble in water and unlike resveratrol (which penetrates the cell passively) the polydatin enters the cell via an active mechanism using glucaose carriers.

Polydatin also exerts cardioprotective effect by the activation of PKC-K(ATP)-dependent signaling and the direct anti-oxidative stress mechanisms. Polydatin affects lipopolysaccharide‘s induction on VEC ICAM-1;Polydatin affects the neutrophils’s chemotaxis reaction inspired by lipopolysaccharide; Polydatin affect smooth muscle and Cardiomyocytes PKC under the situation of ischemia, hypoxia, burn serum, and LPS;

Benefits of taking trans-polydatin supplements

Polydatin is regarded as one of the best supplements in the world for its anti-aging benefits. Polydatin has all the benefits that resveratrol has, such as Anti-aging, Anti-Cancer, cardiovascular support, regulate estrogen level, and weight loss. Polydatin itself is anti-viral, anti-fungal, anti-inflammatory, and has been used in modern China for more than 50 years. Trans-polydatin activates genes which repair and support cell functions, providing many supports for better sleep, more energy, lower sugar, and weight management benefits.
The key benefits of polydatin supplements are listed as below:
1.Polydatin and anticancer;
2.Polydatin has effect on cardiovascular system;
3.Polydatin own the nourishing and protecting liver;
4.Polydatin has the usage of antibacterial and antifungal;
5.Polydatin has impact on the metabolism of bone issue;
6.Polydatin own the effective of antioxidant and quench free-radicals.

Side effects of taking polydatin supplements
Polydatin is a rising ingredient used in dietary supplement, and no adverse effects have been reported yet. As a natural raw material, it is generally regarded as safe. Since it is extracted from polygonum cuspidatum, beware of the purity of polydatin. There may be emodin in the extract which is a laxative and not good to the stomach.
Dosage of polydatin supplements
There is no recommended dosage for pure polydatin (trans-resveratrol-3-O-glucoside). As we know, the dosage of revseratrol98% is about 250mg to 500mg daily. We may conclude that the dose of polydatin is more or less the same as resveratrol dosing. One supplement formula suggests take 1 to 4 capsules of 500mg/capsule a day. High-dose of polydatin show overweight health benefits.
Where to buy high quality polydatin raw ingredient?
Resveratrol series ingredients are Herb Nutritionals Co., Ltd’s main products. There are around 5 resveratrol manufacturers here in China, while only one factory who really manufactures polydatin raw materials since the demand for polydatin is not as huge as resveratrol. Most Chinese export companies are suppliers of polydatin, and their prices are relatively high. We have readily stock of trans-polydatin 98% 200kg each month, certificate of analysis, MSDS, and other documents are available for your reference.
